Alzheimer's risk associated with human apolipoprotein E, alpha-2 macroglobulin and lipoprotein receptor related protein polymorphisms: absence of genetic interactions, and modulation by gender

Abstract

Apolipoprotein E (apoE), the lipoprotein receptor related protein (LRP) and alpha-2 macroglobulin (α2M) have been proposed as a functional complex involved in amyloid clearance, a crucial event for Alzheimer's disease development. In this work, we present an epidemiological approach aimed to study the interactions among these genes, age and gender. This approach did not reveal significant associations between the genes; however, the present study indicated that the risk associated with APOE promoter and LRP gene polymorphisms is modulated by gender.

    These previous studies have demonstrated that minor alleles in this promoter polymorphism are associated with lower AD risk and lower brain beta amyloid deposition (Myllykangas et al., 2002) as well as smaller aging-related change in cognitive function in presence of risk factors such as herpes virus or low attained education (Strandberg et al., 2005). Regarding rs440446, previous studies which have found associations with AD, independent of APOE major isoforms, have most often linked the minor allele with lower risk regardless of the population studied (Bullido et al., 2000; Lambert et al., 1998b; Myllykangas et al., 2002). Our results are also compatible with this pattern.

    This study shows that HNE-bound LRP1 β-subunit, containing the transmembrane portion of the protein, is significantly increased in AD hippocampus compared to age-matched controls, consistent with the hypothesis that oxidative modification to LRP1 contributes to increased Aβ load in AD brain. LRP1 in other tissues is readily oxidized, with resulting loss of function [41,42]. As noted, previous studies show that oxidative modifications to biomolecules occur in AD brain [8,30,43–45].

    The results of a study in a North American white population (Luedecking-Zimmer et al., 2002) strongly suggested synergy between the presence of APOE4 and several single nucleotide polymorphisms (SNPs) around OLR1 (e.g. SF = 2.6, p = 0.001, for the 3′ UTR TT genotype); however, attempts to replicate the interactions in French (Lambert et al., 2003) and Southern Italian samples (D’Introno et al., 2005) have failed. The highly synergistic interaction between APOE4 and the LRP1 exon 3 CC genotype (SF = 6.6, p = 0.007) in a Japanese study (Hatanaka et al., 2000) was not found in a Spanish (Bullido et al., 2000) or a Northern Irish study (McIlroy et al., 2001); it is not known whether this contrast is due to ethnic differences. Two studies have reported interactions between CETP and APOE4: a Spanish study (Rodríguez et al., 2006a), which found synergy between APOE4 and the −629C allele of CETP (SF = 3.1, p = 0.01), but no interaction with the Ile405Val SNP; and a Dutch study (Arias-Vásquez et al., 2007), which showed antagonism between APOE4 and the CETP 405Val allele (SF = 0.66, p = 0.03).
